Effect of Interleukin on the Pulmonary Neutrophil Sequestration After X-ray Irradiation


Abstract
This study was carried out to investigate ultrastructural changes of the lungs and the pattern of inflammatory cells infiltration induced by localized X-ray irradiation combined with cyclophosphamide administration and the effects of IL-1
pretreatment
on X-ray irradiation. ICR mice were injected with 100mg/kg of cyclophosphamide and irradiated 1000cGy of X-ray to the lung and some of them were pretreated by injection of 10§¶/kg of IL-1 24 hours before X-ray irradiation. The mice were electron
microscopically examined 2, 10 and 30 days after X-ray irradiation by morphometry on the neutrophils infiltrated in the lung. The results were as follows
On electron microscopy, after combined irradiation and administration of cyclophosphamide, formation of cytoplasmic processes and detachment from basement membrane of the endothelium, aggregation of platelets, formation of microthrombi, and
infiltration
of neutrophils were identified. On morphometric study, the ratio of area neutrophils per parenchyma was significantly increased in experimental group than that of control. In IL-1 pretreated group, there was no endothelial detachment and
microthrombi
formation Infiltration of the neutrophils were decreased on morphometric study than the group without pretreatment of IL-1.§¶
According to the above results, it could be concluded that pretreatment of IL-1 may have effects to inhibit endothelial damage and to decrease neutrophilic infiltration in early phase caused by combined X-ray irradiation and cyclophosphamide
administration. Therefor IL-1 can ameliorate the pulmonary damage elicited by X-ray irradiation and administration of cyclophosphamide.
